Generic name: Amlodipine and Atorvastatin
Drug class: Antihyperlipidemic combinations, Miscellaneous antihypertensive combinations
Dosage form: Oral tablet
Route of administration: Oral
Dose:
- Initial dose: 2.5 mg amlodipine and 10 mg atorvastatin once daily
- Maintenance dose: 5 mg amlodipine and 10–20 mg atorvastatin once daily
- Maximum dose: 10 mg amlodipine and 80 mg atorvastatin once daily
Mechanism of action: Amlodipine is a calcium channel blocker that relaxes blood vessels, reducing blood pressure and decreasing the heart’s workload. Atorvastatin is an HMG-CoA reductase inhibitor (statin) that decreases cholesterol production in the liver, lowering LDL cholesterol and triglyceride levels while increasing HDL cholesterol.
Drug usage cases:
- Hypertension (high blood pressure)
- Coronary artery disease
- Angina (chest pain)
- Primary hyperlipidemia
- Heterozygous familial hypercholesterolemia in adults and pediatric patients aged 10 years and older
- Homozygous familial hypercholesterolemia in adults and pediatric patients aged 10 years and older
- Primary dysbetalipoproteinemia
- Hypertriglyceridemia
Drug contraindications:
- Active liver disease or unexplained persistent elevations in liver enzymes
- Hypersensitivity to amlodipine, atorvastatin, or any excipient in the formulation
- Pregnancy
- Breastfeeding
Side effects:
- Edema (swelling), especially in the ankles and legs
- Muscle pain, tenderness, or weakness
- Abdominal pain
- Diarrhea
- Headache
- Nausea
- Fatigue
- Dizziness
- Elevated liver enzymes
- Rash
Warnings:
- Monitor liver function tests before initiating therapy and periodically thereafter
- Discontinue therapy if unexplained muscle pain, tenderness, or weakness occurs, especially if accompanied by fever or malaise
- Use caution in patients with a history of liver disease
- Monitor blood pressure regularly
- Use with caution in patients with severe aortic stenosis due to the risk of symptomatic hypotension
Use during pregnancy or breastfeeding: This medication is contraindicated during pregnancy and breastfeeding. Women of childbearing potential should use effective contraception during treatment. If pregnancy occurs, discontinue the medication and consult a healthcare provider.
