Description
Generic name: lumateperone
Drug class: Atypical antipsychotics
Dosage form: Oral capsule
Route of administration: Oral
Dose: 42 mg once daily; may be adjusted to 21 mg or 10.5 mg once daily based on individual factors
Mechanism of action: Lumateperone modulates neurotransmitter activity in the brain, including dopamine, serotonin, and glutamate, to alleviate symptoms of schizophrenia and bipolar depression.
Drug usage cases:
- Treatment of schizophrenia in adults
- Management of depressive episodes associated with bipolar I or II disorder in adults, as monotherapy or adjunctive therapy with lithium or valproate
Drug contraindications:
- Known hypersensitivity to lumateperone or any components of the formulation
- Use in elderly patients with dementia-related psychosis due to increased risk of death
Side effects:
- Sleepiness
- Dizziness
- Nausea
- Dry mouth
- Weight gain
- Movement disorders, including tardive dyskinesia
- Elevated blood glucose levels
- Elevated cholesterol and triglyceride levels
- Low white blood cell counts
- Orthostatic hypotension (low blood pressure upon standing)
- Seizures
- Impaired ability to operate machinery
Warnings:
- Increased risk of death in elderly patients with dementia-related psychosis
- Increased risk of suicidal thoughts and behaviors in children and young adults up to 24 years of age
- Potential for neuroleptic malignant syndrome, a life-threatening condition
- Risk of tardive dyskinesia, which may be irreversible
- Potential for metabolic changes, including weight gain, hyperglycemia, and dyslipidemia
- Risk of low white blood cell counts, increasing susceptibility to infections
- Potential for orthostatic hypotension, leading to falls and related injuries
- Risk of seizures
- Impaired ability to operate machinery due to drowsiness and dizziness
